Doctoral Thesis / Dissertation from the year 2010 in the subject Chemistry - Bio-chemistry, , language: English, abstract: In this study four plants (Chrozophora hierosolymitana Spreng, Chrysanthemum leucanthemum L., Ephedra gerardiana Wall. ex Stapf and Quercus dilatata L.) collected from different regions of Pakistan were screened to identify any chemotherapeutic agentspresent in them. Seven crude extracts of these plants (leaf, stem and root extracts of C. hierosolymitana, aerial parts of C. leucanthemum, stem and root extracts of E. gerardiana and aerial parts of Q. dilatata) were examined for antimicrobial activity using agar diffusion method and agar tube dilution method, cytotoxicity using brine shrimp assay, antitumor activity using potato disc assay, phytotoxic activity using radish seed bioassayand antioxidant activity by using DPPH radical scavenging assay and free radical induced oxidative DNA damage assay.Two plant extracts of C. hierosolymitana and Q. dilatata showed antibacterial activity. Two plant extracts of E gerardiana and C. leucanthemum showed antifungal activity. Two plant extracts i.e., leaf extract of C. hierosolymitna and root extract of E.gerardiana showed significant brine shrimp cytotoxicity activity (IC50 171.55 to 523.8 ppm). Present research work embodies the different methods of Jatropha propagation by using seed germination, stem cutting and tissue culture methods. The extracts of leaf, stem and roots of Jatropha curcas L. were screened in order to study their effects on plant pathogenic fungi, plant pathogenic bacteria and human pathogenic bacteria. Interestingly antibacterial and antifungal nature of plant extracts have been noted. Regarding biochemical analysis of Jatropha species, the studies have made in the part second of the research work and the results are found to be useful in order to record the presence of certain bioactive chemicals in the plants. In order to achieve a mutant ideal plant the studies were carried out by using different mutagenic agents where results showed that certain mutagens are found to be helpful to achieve early flowering and fruiting in Jatropha curcas L.

The present study was undertaken; to evaluate the response of recurrent selection derived lines by the testing of (C3) derived lines for genetic variability, genetic advance and heritability and to assess the effect of selection on other quantitative traits. The 16 half-sib families were selected from 3rd cycle of recurrent selection were grown for evaluation in randomized block design in rabi 2009 -10 along with check varieties Bhima and A1. The expected genetic advance expressed as per cent of population mean at 10 per cent was high for seed yield per plot (48.44) followed by seed yield per plant (28.88). The expected genetic advance per cent over Bhima at 10 per cent selection intensity was high for seed yield per plot (28.27) followed by seed yield per plant (22.78). In safflower seed yield per plant was positively and significantly correlated with days to maturity, number of capitula per plant, number of seeds per capitulum and 100 seed weight. The experiment was laid down in split plot design with three replications, keeping plant density as main plot factor and bulb size as sub plot factor. The effects were studied by collecting data on bulb sprouting and plant performance, yield components, seed yield and seed quality parameters. Analyses of variance, mean comparison using least significant difference, correlation analysis were made using SAS computer software. Plant density significantly influenced bulb sprouting, umbel size, seed yield per 100m2and germination percentage. The largest umbel size (4.9cm) was obtained from low plant density; where as the smallest (4.4cm) from high plant density. Maximum seed yield per 100m2(1,163,446 seeds) was collected from high density while low density yielded only 416,240 seeds per 100m2. However the quality of seed in terms of germination was highest for seeds obtained from low density. Healthy seeds and propagules are the basic requirement for producing good grains, fruits and vegetables needed for human survival and perpetuation. Dispersal of microbial plant pathogens via seeds and propagules has assumed more importance than other modes of dispersal, as infected seeds and propagules have the potential to become the primary sources of carrying pathogen inoculum for subsequent crops. Several diseases transmitted through seeds and propagules have been shown to have the potential to damage economies as a result of huge quantitative and qualitative losses in numerous crops. Hence, it is essential to rapidly detect, identify and differentiate the microbial plant pathogens present in seeds and propagules precisely and reliably, using sensitive techniques. To compete favourably in this new global seed world, quality and cost are and will certainly continue to be the key issues. The presence of counterfeit seed in Uganda seed market has been reported. Studies need to be conducted to investigate at which level along the seed value chain the deterioration in quality occurs. The current study assessed the change in quality of hybrid maize seed as it is transferred from seed companies (when seed has been processed and packaged) to agro-dealers. Specific objectives of the study were to determine the levels of phenotypic and genotypic variation in hybrid maize seed from seed companies and agro-dealers. A total of 120 samples of four hybrid maize varieties (improved varieties most favored by farmers) used in this study were collected from agro-dealers in 15 districts and from 3 seed companies. Alpha lattice design with two replications has been used. The present investigation was carried out in Randomized Block Design during rabi season of 2005-06 with 25 genotypes derived from wide and intervarietal crosses and 5 checks of chickpea at Crop Research Center of G.B. Pant University of Agriculture and Technology, Pantnagar, Uttarakhand. The observations were recorded on days to 50% flowering, days to maturity, plant height, primary branches/, plant width, pods/ plant, seeds/pod, 100 seed weight and grain yield/plant to study genotypic, phenotypic variability, genetic advance, character association, genetic divergence and genetic relationship using SDS -PAGE. The ANOVA revealed highly significant genetic differences among genotypes for all the characters studied. High values of C.V. were observed for grain yield/plant, 100 seed weight, primary braches/plant and seeds/ pod. Amongst 25 genotypes only PG055 and PG053 were significantly superior for plant width and grain yield/ plant, respectively. Analysis of genotypic character association study revealed highly significant and positive association of days to maturity with plant height (0.496), seeds/ pod and days to 50% flowering (0.491), primary branches/plant and pods/ plant

The studies carried out on the role of honey bee (Apis mellifera) in hybrid seed production of Brassica campestris showed the highest number of pollen and nectar collectors of Apis mellifera and were recorded 45.6 and 24.6 per square meter crop area of B campestris respectively during day hours of 12.00 to 2.00 pm, which were significantly greater than the rest of foraging observations. In bee pollinated plants, the number of pods per plant, number of seeds per pod, number of seeds per plant, seed yield per plant and weight of 1000 seeds in BP plants were about 1.3, 1.1, 1.44, 1.53 and 1.62 times greater than NP plants,respectively. As such worker bees observed for foraging to collect the nectar and pollen after spraying of the insecticide i.e. Confidor 200 SL (Imidacloprid) in the concentration of 0.025g a.i., 0.030g a.i.and 0.035g a.i., but bee poisonong were neither observed in bee hive nor in field. During foraging, honey bees provide the opportunity for transference of pollen from male fertile to male sterile plant and resulted the production of hybrid seeds.

Scientific Study from the year 2012 in the subject Biology - Micro- and Molecular Biology, Nirma University (Institute of Science), language: English, abstract: Extracts of five different plant seeds- Syzygium cumini, Pheonix sylvestris, Manilkara zapota, Tamarindus indica, and Annona squamosa- prepared by Microwave Assisted Extraction (MAE) method were screened for their antimicrobial activity against certain phytopathogenic microorganisms. Ethanolic extract of S. cumini was found to possess highest average total activity against susceptible microbes. Total activity was found to have a positive correlation with the extraction efficiency. Methanolic extract of T. indica exerted bactericidal action against Agrobacterium tumefaciens and Pseudomonas syringae. Ethanolic extract of P. sylvestris was able to protect cabbage leaf against Xanthomonas campestris. Acetone extract of M. zapota was able to reduce aflatoxin production in Aspergillus parasiticus by >50 %. Curcumin proved bactericidal against X. campestris.

The Bt (Bacillus Thuringiensis) cotton seed production focussed on the socio-economic characteristics of contract farmers, cost and return, efficiency of resource use and problems faced by the farmers in Bt cotton seed production under different companies. The total cost of Bt cotton seed production varied from one company contract farmers to other company contract farmers. The Cobb-Douglas production function revealed that the farmers who have contracted with different seed companies, the use of seed was found to be optimum across all company contract farmers. Whereas, fertilizer resource was over utilized by farmers who have contracted with various companies. Non availability of trained labour and prevalence of high wage rate were the major constraints in case of production, where as poor technical assistance and low contract price were the contractual problems. Therefore the Bt cotton seed production technology had positive impact on Socio- economic status of farmers by increase in yield and reducing cost on inputs thereby increase in income and also standard of living.

Mikania scandens(L.)Willd. is a rapidly growing herbaceous hemp vine belongs to the family Asteraceae. Present study is concentrated on anthelmintic, antimicrobial and phytochemical aspects on Mikania scandens(L.)Willd. The phytochemicals of leaves of Mikania scandens(L.)Willd. was extracted using polar and non polar solvents viz, Hexane and Methanol. The antimicrobial activity of extracts were tested against Gram- positive and Gram- negative bacteria and two fungal strain. The essential oil also screened against test strains and show better antimicrobial activity. In vitro experiment was performed to evaluate anthelmintic activity of extracts, using Pheretima posthuma. The polar solvent shows better activity than the standard Albendazole. The methanol extracts of Mikania scandens(L.)Willd. nine compounds were analyzed by HPLC. The essential oil is obtained by hydrodistillation of the whole plant of Mikania scandens , fifteen compounds were analyzed by GC-MS. Various kinds of monoterpenoids, diterpenoids and sesquiterpenoids were the major phytochemicals detected.

Under favourable conditions such patches may increase in size until they merge with each other and a closed canopy forest builds up. However, in dry areas successive invasion into grassland is blocked and there is little spread outwards.Research was conducted from September 2005 to January 2006 in the Western Soutpansberg, South Africa. For Mimusops zeyheri and Syzygium legatii effects of shade and seed size on germination and seedling recruitment have been investigated in plant pot experiments. Two field experiments were conducted investigating seed predation and the effect of competition. Additionally, recruitment was studied at eight tree islands and soil samples from 96 plots varying in exposition, habitat type and location of sampling site were analysed.Results revealed that tree recruitment was influenced by complex interactions between facilitation and competition, herbivory and abiotic environmental stress. In this laboratory investigation, the possibilities of using unconventional feedstuffs for poultry from animal origin as well as plant origin were studied. The selected unconventional feed resources used in this study may fulfill the nutritional requirement of poultry industry. Animal origin of unconventional feedstuffs (e.g. blood meal, feather meal, leather shavings, silkworm pupae, poultry droppings, poultry offal meal, poultry manure and poultry hatchery byproduct) contain higher % of protein than the plant origin of unconventional feed resources (e.g. drumstick leaf meal, krishnachura leaf meal, ipil ipil leaf meal, duckweed meal, waste tea leaf meal, rubber seed meal, neem leaf and seed meal, cassava leaf and stem meal, bitter melon seed meal and triticale meal). As the selected samples are easily available, these can be collected and processed easily for the use and may minimize the cost. Therefore, it is concluded that the unconventional feed samples may be an efficient and economic substitute of conventional feeds. It is possible to replace conventional feeds by cheaper unconventional feeds in poultry diet leading to increase profitability.

It is estimated that ca. 16% of the world's agro-climatic areas are affected by waterlogging. It causes significant reduction in the yield of several crops. Jatropha curcas L. is a deciduous shrub, drought-resistant but prone to waterlogging, oil bearing plant species, widely distributed in tropical and sub-tropical areas viz., in Central and South America, Africa, India and Southeast Asia. The plant produces useful products particularly seeds from which oil can be extracted (~35%), may be used in soap and cosmetics industries, and also as an important substitute for diesel and kerosene. Accordingly, it is expected that by 2015, Jatropha may be planted on 12.8 mha land globally for blending of fossil diesel with biodiesel across the world under climate change to mitigate green house gases (GHGs). An improved carbon assimilation through photosynthesis to achieve enhanced net plant produce and oil content is the need of future. Charcoal rot of mungbean caused by Macrophomina phaseolina is a destructive disease of mungbean. Nowadays, scientists are in search of alternative environmental friendly disease management strategies to reduce the negative effects of synthetic agrochemicals on environment. The present study was designed to control the pathogen and the disease by using different species of a biological control agent Trichoderma, and extracts and dry biomass of Sisymbrium irio, a weed of family Brassicaceae. Root and leaf extracts of the weed showed the best inhibitory effect on growth of M. phaseolina in laboratory bioassays. among the seven species of Trichoderma, T. harzianum showed the best antagonistic activity against the pathogen. In pot trials, T. harzianum in combination with 1% dry leaf biomass of S. irio as soil amendment enhanced plant resistance against the fungal pathogen and significantly increased grain yield of mungbean by 62% and 805% over negative and positive control treatments, respectively. Healthy seeds and propagules are the basic requirement for producing good grains, fruits and vegetables needed for human survival and perpetuation. Dispersal of microbial plant pathogens via seeds and propagules has assumed more importance than other modes of dispersal, as infected seeds and propagules have the potential to become the primary sources of carrying pathogen inoculum for subsequent crops. Several diseases transmitted through seeds and propagules have been shown to have the potential to damage economies as a result of huge quantitative and qualitative losses in numerous crops. Hence, it is essential to rapidly detect, identify and differentiate the microbial plant pathogens present in seeds and propagules precisely and reliably, using sensitive techniques. Microbial Plant Pathogens: Detection and Management in Seeds and Propagules provides a comprehensive resource on seed-borne and propagule-borne pathogens. Information on the biology of microbial pathogens, including genetic diversity, infection process and survival mechanisms of pathogens and epidemiology of diseases caused by them, are discussed critically and in detail to highlight weak links in the life cycles of the pathogens. Development of effective disease management systems, based on the principles of exclusion and eradication of pathogens and immunization of crop plants to enhance the levels of resistance of cultivars to diseases, has been effective to keep the pathogens at bay. The need for production of disease-free seeds/propagules has been emphasized to prevent the carryover of the inoculum to the next crop or introduction of the pathogens to other locations. Effectiveness of adopting simple cultural practices and development of cultivars resistant to diseases through traditional breeding methods or biotechnological approach have resulted in reducing the pathogen inoculum and disease incidence. Although application of different chemicals may reduce the disease incidence effectively, biological management of crop diseases, employing potential biological control agents have to be preferred to preserve the agroecosystems. Greater efforts have to be made to integrate compatible strategies to enhance the effectiveness of diseases management systems.
