Of the handful of neurotransmitters modulating taste response, norepinephrine (NE) may be the least understood. Although the physiological role of NE has been tested, it is still unclear whether the rat taste bud may serve as an endogenous source of neurotransmitter or if it must be taken up from outside the taste bud. Immunocytochemical analysis of DBH has shown that DBH-LIR is present in taste cells of the rat circumvallate papillae. Specifically, DBH-LIR is present in a subset of Type II taste cells. Nearly all (98%) DBH-LIR cells express PLCβ2-LIR, and about half (41%) of PLCβ2-LIR cells express DBH-LIR. DBH-LIR does not co-localize with NCAM, and thus, is not present in Type III cells. NET-LIR is present in the taste cells of the rat circumvallate papillae. Double labeling of NET-LIR and IP3R3-LIR indicates that NET is present in Type II taste cells. A large proportion (77%) of NET-LIR cells express IP3R3-LIR and all (100%) IP3R3-LIR cells express NET-LIR. A small proportion (7.8%) of NET-LIR cells also express syntaxin-1-LIR in Type III cells. About a third (32.8%) of syntaxin-1-LIR cells express NET-LIR.

Multiple myeloma (MM) is a malignancy of plasma cells (PCs) which accumulate in the bone marrow (BM), thus, leading to anemia, bone lesions, hypercalcemia, renal impairment and immune dysfunction. T regulatory (Treg) cells maintain immune homeostasis in healthy individuals by patrolling unwanted immune responses. Whereas in cancer patients, these cells are uncontrollably expanded, thereby, inhibiting the host anti-tumor responses. Majorly this work is focused to understand the role of Treg cells, including CD4 Treg cells and CD8 Treg cells in rendering the immune impairments in MM patients. This study clearly showed that both CD4 and CD8 Treg cells were functionally suppressive and increased in MM patients. These findings were confirmed by using different methods, including flow cytometry, functional and molecular studies (RT-PCR and western blot). Clinical significance of this study was CD4 Treg cells were found to be increased in patients with shorter time to progression, hypercalcemia, lower normal PC counts (≤ 5%) and IgA myeloma subtype. From these findings, Treg cells could be considered as one of the evasion mechanisms of myeloma cells from host immune responses.

Hematopoiesis is maintained throughout the lifetime of an organism by a specialized group of rare cells known as the hematopoietic stem cells (HSCs). These cells are defined by their ability to self-renew to give rise to similar daughter HSCs, and to differentiate into all the lineages of blood cells. The region where HSCs reside to fulfil these tasks is known as the stem cell niche. In a niche, neighboring subsets of cells and extracellular substrates house stem cells and provide specialized signals to modulate stem cell self renewal and progeny production. Many studies have identified key components of the niche required to maintain the HSCs in their primitive state. However, the exact location of the HSC niche is yet to be clearly identified. Understanding all of the interactions between the stem cells and their microenvironment may yield practical methods for manipulating stem cells to achieve therapeutic outcomes in various disease settings.

Urothelial cells can be harvested for cell expansion outside the body with a minimally invasive technique. The cells can be stored for later transplantation back to the patient. In this procedure the urinary bladder is catheterized and washed with sterile saline for isolation of cells. The method can be used for adult and pediatric patients. In a clinical study, we used in vitro cultured autologous cells for urethral reconstruction in patients with severe hypospadias. Follow-up studies demonstrate good results. Our laboratory work includes quality controls of urothelial cells after long-term expansion in vitro and an optimized cell culture protocol. We also demonstrate that three different cell types can be co-cultured to achieve a complex tissue engineered transplant that mimics the wall of the urinary pathway.

In contrast to most other vertebrates,teleost fish exhibit an unparalleled capacity to generate stem cells in both the intact and the injured adult brain.My research is focused on the occurrence, the extent and the fate of aneuploid cells (genome-modified individual cells characterized by the loss or gain of whole chromosomes) generated in the teleostean brain. Aneuploidy arises through various chromosome segregation defects during the mitotic divisions of newly-generated cells in the adult brain. Aneuploidy is observed in ~80% of the newly-generated stem cells of the adult brain,but occurs at a very low rate in liver tissue, as shown by metaphase chromosome spreads and flow-cytometric DNA content analysis. The mitotic machinery of adult brain cells was examined for chromosome segregation defects which showed lagging chromosomes at metaphase and anaphase, as well as micronuclei and anaphase bridges,in the intact adult brain. In conclusion,my research demonstrates the evidence of the generation and maintenance of aneuploid cells in the adult fish brain that arise through chromosome missegregation which is a widespread phenomenon in the teleostean central nervous system.

Bone Tissue Engineering is a promising field that helps human needs by applying the principles of biology and engineering to the development of feasible substitutes that restore and retain the function of human bone tissues. The live cells in the body attach to the normal scaffold known as extracellular matrix but in in-vitro studies presence of a scaffold is essential to keep cells alive. Therefore, good cell adhesion on scaffolds is essential for successful tissue engineering. The aim of this book is to identify the key factors of coating which leads to better cell adhesion and to establish the relationship of cells’ adhesion and the treatment of substrates. Another aim is to apply four-point bending system to stimulate bone cells and finally evaluate bone cells’ response to the mechanical stimuli on different substrates by morphology alteration using histological and immunological staining methods. Human bone cell line (MG-63) and rat primary bone cells were used in this study. Two-dimensional substrates were used as a scaffold model. The selected substrates included PLLA, PLLA coated with PDLLA-b-PEO co-polymer, collagen coated, RGD grafted PLLA-PEO coated substrates.

Within this work electrochemical processes for manufacturing of novel silicon solar cells are investigated. Direct plating of Ni and Al on n- and p- silicon is demonstrated by making use of solar cell characteristics. Homogenous Ni/Cu stacks are realized for bifacial and back contact solar cells, forming an excellent mechanical and electrical contact to silicon. For metallization of HIT solar cells, the plating behavior on ITO layers is studied. Additionally, plating processes on evaporated Al layers are developed and applied to back contact solar cells. By means of process optimization the plated metal stack on Al features sufficient adhesion and increases the lateral conductivity of the metal grid resulting in increased solar cell efficiency. An advanced metallization route for back contact solar cells which purposefully utilizes the different characteristics of the deposited metals (Al, Ni, Cu) is developed. The resulting metal stacks are characterized in detail using SEM, EDX and AES methods. Besides plating processes, local oxidizing processes for Al are established and combined with printing technologies to realize the metal contact separation for back contact solar cells.

This has unlocked a new direction of research to restore the form and function of the oral cavity using autologous cells, thereby preventing histocompatibility mismatch and transmission of disease. With the first reports of adult human stem cell populations residing in the periodontal ligament beginning to emerge, the next phase will be to determine the clinical utility of these cells. Accordingly, further studies are now required to determine the efficacy of ex vivo expanded stem cells to repair periodontal defects.

High operation temperature fuel cells allow internal reforming, promote rapid kinetics with non-precious materials and offer high flexibilities in fuel choice, and are potential and viable candidate to moderate the fast increase in power requirements and to minimize the impact of the increased power consumption on the environment.
