Fast dissolving film can be defined as a dosage form, which when placed in the oral cavity it rapidly disintegrates and dissolves to release the medication for oral mucosal absorption or allow for the gastrointestinal absorption to be achieved when swallowed. Ketotifen Fumarate (KF), antiasthmatic and antiallergic has oral bioavailability of 50% due to hepatic first pass metabolism. This study aims to formulate ketotifen fumarate as oral dissolving films, to improve the bioavailability by avoiding hepatic first-pass metabolism. Nineteen formulas were prepared using solvent-casting method, and the effect of different formulation variables on the physical and mechanical properties of the prepared films, besides to the drug release behavior was evaluated. The prepared oral film of ketotifen fumarate that contains HPMC (6cp) showed the fastest in- vivo/in- vitro disintegration time among other investigated polymers. The results also showed that as the concentration of HPMC decreased, both the disintegration and the drug release rates increased, it was also seen that the disintegration and the drug release rate increased significantly as the concentration of tween 80 is increased

For the enteric layer, influence of polymers and solvent system was preventing the drug release in the stomach environment with targeting at duodenal ulcers. The main focus of this research work was to develop a matrix type transdermal patch containing Propranolol hydrochloride and combination of two hydrophobic Polymers Eudragit L-100 and Eudragit S-100 by Solvent Evaporation technique.Combination of two Plasticizers Glycerine and Dibutyl phthalate were used and their effect on drug release was studied by changing their concentration in each batch.Methanol and Dichloromethane solvent system was used in which methanol acts as a penetration enhancer.Since oral bioavaibility of Propranolol Hydrochloride is poor due to high first pass metabolism transdermal patch is formed with the objective to get sustained release drug action with good bioavaibility. It can be concluded that the attempt of formulation and evaluation of the propranolol hydrochloride patches was found to be successful in the release of drug for an extended period of time. Further detailed investigations and elaborate in-vivo studies need to be carried out and an in-vitro in-vivo correlation need to established to guarantee the efficiency and bioavaibility of the formulation.

Scientific Study from the year 2014 in the subject Biology - Micro- and Molecular Biology, Tamil Nadu Agricultural University, language: English, abstract: Root knot nematode, Meloidogyne incognita, is an important nematode parasite of tuberose. A study was undertaken to assess the evaluation of plant growth promoting rhizobacteria (PGPR) against Meloidogyne incognita infestation on tuberose in vitro, pot culture and field conditions. Tuberose fields were surveyed for the nematode incidence in different districts of Tamil Nadu. Among the nematodes associated with the crop M. incognita caused severe damage to crop. Plant growth promoting rhizobacteria were isolated from soil samples collected from field and subcultured for further studies. The isolates were tested for their efficacy for plant growth promotion by roll towel and pot culture studies with rice plants. Among them, three isolates, viz. BT3, BT8 and BT21 were found to be effective in increasing the plant growth promotion when compared to other isolates. Biochemical characterization of the isolates revealed that the cultures were found to be the group of Bacillus spp. Liquid formulation were prepared for the three isolates with nutrient amendments, viz. glycerol (10 Mm), trehalose (5 mM) and the stickers, viz. starch (2%) and PVP (2%) and the antagonistic effect of different Bacillus isolates were studied against M. incognita in vitro. Niosomes was developed and characterized for ocular drug delivery of diclofenac potassium. The present research study is a promising approach to improve corneal penetration and bio availability characteristics. Formulation also found to ensure a good entrapment efficiency and ocular bio availability of drug in-vivo. Niosome containing diclofenac potassium were prepared using surfactant and cholesterol in different ratio by Lipid film hydration technique. Niosomes were characterized For Entrapment efficiency, Particle size analysis, In-vitro drug release and In-vivo studies. Formulation with low cholesterol content which shown 82.1% Entrapment efficiency, 70.01% sustained release over a period of 10 h followed a non-fickian profile with zero order release profile. Scanning electron micrograph indicated that niosomes have a discrete spherical structure without aggregation. In-vivo study showed an availability of drug in aqueous humor for an extended time period even up to 8 hour and it showed a correlation with the release profile in-vitro. Niosome are considered the best as it showed good and high Entrapment efficiency and Vitro release with better bio availability.

The aim of this research was to formulate and evaluate in-situ liquid depot from bio-degradable polymers like PLGA 85:15 and PLGA 50:50.Isoniazid is a drug of choice for the treatment of tuberclosis. It requires prolong therapy, it has very short half life (Fast acetylators) of 1.1 + 0.1 to 2-3 hrs. Isoniazid is readily absorbed orally and penetrates all body tissues; it is extensively metabolized in liver, most important pathway being acetylation metabolites are excreted in urine. The developed Isoniazid Insitu liquid depot was characterized for percent drug content (PDC), drug polymer interaction study, viscosity, and drug release study. The results of the study showed there were no interaction in drug and polymers after formulation, viscosity was found in all formulation between 1.02 to 1.57 poise and drug content was found to be very between 92.39 + 0.47% to 97.15 + 0.31%. It was found that the amount of drug released at after 168 hours was greater from PLGA 85:15 depots systems formulations as compared to PLGA 50:50 biodegradable liquid depot injections. The results of the characterization and evaluation of in-situ liquid depot established the safety for use and suitability.

The book contains details of study carried out to evaluate the effect of various penentration enhancers (PE) on the transdermal permeation of dexamethasone. In vitro studies were performed in order to determine the optimum solvent system for the drug. The concentration of gelling agent HPMC was optimized. Further formulations were prepared and optimized for the concentration of PE to achieve maximum permeation of drug. In vitro skin permeation studies were performed using selected formulation for 24 hours, which revealed that formulation containing 5% w/w eucalyptus oil and camphor oil showed cumulative drug permeation in the desired range. Based on the observation it can be concluded that volatile oils containing terpenes are better studied over alcohols and fatty acids for increasing the permeation of dexamethasone. The formulation containing terpenes could be useful in the development of transdermal drug delivery system of dexamethasone

Oral route is preferred for the treatment of various diseases/disorders. However, the oral bioavailability of nearly 40% of newly discovered molecules is diminished due to various physicochemical and physiological barriers. The reasons of low and variable oral bioavailability include, high lipophilicity, restricted permeability, first pass metabolism in the gut and liver and efflux transporter systems. These problems, in turn, lead to high intra- and inter-subject variability and lack of dose proportionality. To overcome these drawbacks, various formulation strategies have been adopted. However, the focus has recently shifted to lipid-based formulations owing to their ability to improve the oral bioavailability of poorly water soluble drug compounds. In this regard, self-emulsifying drug delivery systems (SEDDS), particularly nano-based systems, have been able to surmount the above mentioned problems. SEDDS are the recent technological innovations which are physically more stable and easier to manufacture with the globule size ranging between 10 to 1000 nm. Not only this, SEDDS are able to modulate the drug absorption leading ultimately to enhance oral bioavailability.

This study is carried out to design controlled release multilayered matrix tablets of antihypertensive drug. The aim of this research work is to reduce the frequency of dosing, decreasing the required dose employed, providing uniform drug delivery, reduction of side effects and enhancement of patient compliance of antihypertensive drug. The purpose of study to formulate and evaluate multi-layer matrix tablets using hydrophilic polymer, namely xanthan gum and chitosan by the wet granulation. Captopril was choosen as a model drug because of its higher water solubility. From the results obtained, the hydrophilic polymer xanthan gum and chitosan used individually could not sufficiently produce controlled release so must be combined in various ratios for effective oral controlled release to be achieved of multilayer matrix tablet that complied with all parameters. All batches were found to have almost three months stability in terms of globule size and phase separation.

Most of herbal formulations have lacking of preformulational and post formulational studies. Thus, an attempt was made to performed the preformulation and post formulation characteristics for senna leaf powder and calcium sennoside. The conventional tablet of senna leaf powder and calcium sennoside may release a significant amount of drug in the physiological environment of stomach before reaching at the site of action. Hence, enteric coated tablet of calcium sennoside and senna leaf powder was prepared using cellulose acetate phthalate for averting the initial loss of 5-8% sennoside in stomach. For this purpose different formulational factors were optimized. The in-vitro release of coated tablet was performed in 0.1 N Hcl and phosphate buffer of pH 6.8. After the enteric coating, the tablet ensured that it was more pH dependent and release the drug only at higher pH. This research work should be especially useful to professionals engaged in formulation development of herbal drugs, or anyone else who wants to perform the preformulation and post formulation study of herbal drugs.

Fast dissolving films have become popular as a new delivery system because they are easy to administer and sudden onset of drug action is possible as the films are taken through the sublingual route. In present study Zolmitriptan fast dissolving sublingual films were prepared which allow fast, reproducible drug dissolution in the oral cavity, thus bypassing first pass metabolism to provide rapid onset of drug action. The fast dissolving films were prepared by solvent casting method. Low viscosity grade of hydroxylpropyl methylcellulose (HPMC E5) and maltodextrin were used as film forming polymer due to their hydrophilic nature. Proposed combination provides acceptable dissolving criteria owing to HPMC E5 and better mechanical properties due to maltodextrin. Propylene glycol, citric acid, mannitol and mango flavour were used as a plasticizer, saliva stimulating agent, sweetener and flavouring agent respectively. Drug-excipients compatibility study was done using FT-IR spectroscopy. The prepared films were evaluated for thickness, weight variation, disintegration time, surface pH, folding endurance, drug content, in vitro dissolution, tensile strength and % elongation.

Despite advances in surgery, radiotherapy, and chemotherapy, the survival of patients with oral squamous cell carcinoma has not significantly improved over the past several decades. A potential approach to the treatment of oral premalignant and malignant disorders is gene therapy.
