Plant growth regulating fungi are associated with plant roots and secrete a number of useful secondary metabolites in the rhizosphere. We investigated plant growth regulating activity of roots inhabiting endophytic fungi isolated from different weed species. Endophytic fungi were isolated from Echinochloa crus-galli, Digitaria sanguinalis, Monochoria vaginalis, Plantago lanceolata, Portulaca oleracea, Veronica arvensis, and one vegetable Cucumis sativus. We isolated 189 fungal endophytes from the roots of these plants, of which 78 fungal isolates were screened for their effects on plant growth and development. Giberellins production by selected fungal endophytes were also studied.

This thesis summarizes the microbial transformation of two compounds, Estrone, a steroid and Cedryl Acetate, a terpene. The steroid, estrone (1) was transformed by two microbes, Cunninghamella elegans and Rhizopous stolonifer. When transformed by Cunninghamella elegans, two transformed products were produced including metabolites (2) and (3), one showed hydroxyl addition while other showed addition of hydroxyl group along with conversion of carbonyl group to hydroxyl group. Estrone also gave transformed products by Rhizopous stolonifer which are similar to that obtained by Cunninghamella elegans. The terpene, cedryl acetate (4) when transformed by Cunninghamella elegans gave three transformed products includes metabolites (5), (6) and (7), showed addition of hydroxyl group at different positions.

Wood decay is a deterioration of wood by primarily enzymatic activities of microorganisms. For practical purposes, fungi are the only agents of wood decay. Fungi which grow on wood are sometimes called "lignicolous" fungi. But why develop a set of keys limited to fungi utilizing wood as a substrate? After all, being lignicolous does not define a taxonomic category. Lignicolous fungi include ascomycetes and basidiomycetes and a large number of classes and orders within each of these groups. Most of these taxa include both lignicolous and terrestrial species. Rather than taxonomy, the keys focus on the biological activity holding this otherwise disparate group of fungi together: their ability to degrade cellulose and lignin, the major components of wood. Forest trees and valuable landscape trees can be infected and rotted by these fungi. Knowing the species growing on a tree can help the forester determine the likely extent of loss. Different species are associated with different amounts of decay in the tree. Additionally, some species are restricted to sapwood and will not affect the merchantable volume of heartwood. Some fungi can decay sound wood; others decay only decaying wood.

The ergogenic potential of r-HuEPO, demonstrated through improved aerobic capacity and performance has reported r-HuEPO to be the most widely abused erythropoietic stimulant till date. This study was aimed at determining the effects of r-HuEPO on the human metabolome by analyzing metabolites from blood plasma and urine. Analysis of these metabolites was then used to determine how such metabolic interactions affected physiological status and exercise performance. Three well trained individuals participated in the study. Blood and urine were collected from the subjects. Plasma and urine samples were analyzed for metabolites by LC-MS Orbitrap and data was analyzed in the form of heatmaps and PCA plots. Further data interpretations for metabolites of interest were performed by a software, mzMatch/PeakML. Analysis of the human metabolome revealed 1000 metabolites which were manually categorized into 190 human metabolites. Trichothecenes are mycotoxins produced by Fusarium fungi commonly found in cereals. The toxins have been associated with diseases in humans and farm animals. Trichothecenes are chemically closely related toxins with an epoxide ring. The micro- organisms in faeces from pigs had the ability to transform trichothecenes to de-epoxy metabolites. The ability was transferred between pigs when faeces were transferred between pens. Human faeces were not able to transform trichothecenes. The de-epoxides were shown to be less cytotoxic than toxins with an intact epoxide ring, verifying that the transformation is a detoxification. 3-acetyl-DON was rapidly de-acetylated and absorbed in pigs followed by a rapid excretion. No accumulation occurred in plasma. No de-epoxy DON was present in plasma or urine, indicating that the de- epoxidation occurs in the lower parts of the gastrointestinal tract where the absorption is low. DON and conjugated DON were the only metabolites detected in plasma and urine. Feeding studies with DON and T-2 given in feed to farm animals were reviewed and guideline limits for feed proposed.

Mass screening for colorectal cancer (CRC) and removal of precursor lesions dramatically reduces both incidence and mortality from CRC. Most of the eligible U.S. population has not been screened. Disparity of screening exists within populations across the domains of race/ethnicity, gender and age. This research describes the patients eligible for colorectal cancer screening and the practice of provider ordering and patient refusal of colorectal cancer screening in one Veteran’s Affairs Healthcare setting (VA). This is a descriptive cross sectional study utilizing secondary analysis of data from VA electronic medical record system. 4,315 men and women aged 50 years and older eligible for screening between 2004 - 2005. Independent variables: demographic characteristics age, gender, race. Dependent variables: provider order, type of screening, screening not indicated, patient refusal of screening. Conclusions: Providers determined screening was not indicated primarily due to patient chronic health problems. A significant number of patients refused provider recommendations for screening. The reasons for patient refusal are unclear. Young black men refused more than any other group.

The study includes screening by morphological characters and subsequently validation using SSR (Simple Sequence Repaet) markers. Most importantly this research will indirectly ensure food security.

Methylmalonic acidemia (MMA) is an autosomal recessive disorder of methylmalonate and cobalamin (cbl; vitamin B12) metabolism. It results commonly from a defect in the gene encoding methylmalonyl CoA mutase apoenzyme MCM (mut MMA or vitamin B12-unresponsive MMA). This work highlights four aspects of methylmalonic aciduria diagnosis: First, it presents an overview of the diagnostic evaluation of patients with organic acidurias and several other IEMs by liquid chromatography- tandem mass spectrometry (LC-MS/MS), gas chromatography- mass spectrometry (GC-MS) and isocratic “high-performance liquid-chromatography” (HPLC).Second, it reports HPLC method for screening of disease associated metabolites and compares the specificity and sensitivity of HPLC and GC/MS for organic aciduria screening. Third, it reports the clinical and biochemical laboratory investigations to MMA, and finally was to highlight the molecular investigations by reporting the results of mutation study of exon II of MUT gene for all patients with methylmalonic aciduria. Findings are likely to be of great interest to the scientists, researchers, trainees and clinicians and could be applied in the clinic right away.

Plants synthesize certain chemicals for the sake of protection against the organisms which are harmful to them or for the sake of competition for the natural resources. These chemicals are generally categorized as secondary metabolites. The secondary metabolites are equally beneficial for human being too as these are medicinally important. Literature survey shows the enhancement of plumbagin content was enhanced by micro-propagation through hairy root culture. The plumbagin was also found to be increased through cell culture technique by elicitation. In the present investigation attempts have been made to enhance plumbagin content in Plumbago zeylanica by entirely different method i.e. in vivo elicitation through bacterial treatment.

The evidence we reviewed indicates that a reasonable age at which to initiate cervical cancer screening in women is age 21. Screening before this age is complicated by relatively high rates of transient HPV and regressive cervical abnormalities, with very few actual cancer cases. Current data cannot assure that beginning screening after this age is clearly safe, particularly in the United States, which has no centralized national cervical cancer screening program. For cytology-based screening, LBC does not differ from CC in sensitivity, specificity, or relative CIN detection, but may yield a lower proportion of unsatisfactory slides. Cost, overall screening strategy, and other considerations may also pertain to local decisions on which approach to use for collecting cytology samples. In women older than age 30 years, a single HC2 test is clearly more sensitive for CIN2+ and CIN3+ (about 40% greater) than cytology alone. However, a single HC2 test is also 3 to 5 percent less specific than cytology.

The results of phytochemical screening indicated the presence of secondary metabolites such as tannins, betacyanins, carbohydrates, alkaloids, terpenoids, phenols, quinines, saponins, cardiac glycosides etc. Also the comparative antimicrobial activity of chloroform and water extracts of fruit, leaf and stem bark of Annona reticulata was evaluated against four bacterial species namely, Escherichia coli, Pseudomonas aeruginosa, Serratia marcescens and Micrococcus luteus and two fungal species namely Candida albicans and Rhizopus. Agar well diffusion method and disc diffusion method were selected to check the antimicrobial activities of the extracts. The study revealed that the chloroform extracts of leaf, stem bark and fruit of Annona reticulata has activity against the bacterial strains and fungal strains. During this time, I worked closely with the local school nurses and witnessed first-hand the many challenges these health workers contend with. Through my collaborative work with the school nurses I identified an urgent need for a contextually relevant hearing screening protocol to assist in improving the current state of the screening service. There is evidence in support of the conduct of routine hearing screening in our schools but limited research has been done to translate the theory into a reality. This study served to address this shortcoming using a collaborative approach in which the school nurses had a definite voice, contributing to the selection of the most appropriate hearing screening protocol for use in a school setting.
