Onasanya Amos Proteomic Diversity in Xanthomonas Oryzae Pv. Oryzae from West Africa

Proteomic is now increasingly considered as useful technique to reveal genetic diversity among different bacteria isolates differentiating live pathogens from dead ones. This book, therefore, reveals proteomic diversity of thirty Xanthomonas oryzae pv. oryzae (Xoo) isolates, the causative agent of rice bacterial leaf blight disease in West Africa, using isozyme polyacrylamide gel electrophoresis (PAGE) analysis. The study revealed 23 isozyme loci in which SKDH produced 33.3-93.3 % polymorphism, EST and G6PH equally gave 40-96.7 % polymorphism within the Xoo isolates enzyme profile. Phylogenetic relationship among 30 Xoo isolates revealed two major genetic groups (Xoo-A and Xoo-B) with two subgroups each (Xoo-A1 and Xoo-A2) and (Xoo-B1 and Xoo-B2). The distinct pattern of each isolate suggested high level of genetic variation and frequent occurrence of mutants in Xoo isolates in different host cells. This information should especially be useful for rice breeders, rice pathologists, rice disease epidemiologists, rice biotechnologist and the scientific community aiming at development of durable Xoo resistant rice cultivars to different localities in West Africa.

Bachelor Thesis from the year 2012 in the subject Biology - Micro- and Molecular Biology, grade: A pass, , course: Molecular Biotechnology, language: English, abstract: Traditional rice varieties consists of many important traits such as medicinal and nutritional value, drought and pest resistance. Efficient tissue culture and transformation techniques are important in uncovering such genes from traditional rice varieties. Aim of this study was to optimize an efficient protocol of callus induction and transformation of Sri Lankan traditional rice varieties. Dahanala, Kaluheenaty, Kalubalawee, madael, Kahatawalu, Kaharamana, Gonabaru, Alwee, Wannidahanala, Kottiyaran, BG380 and H4 were selected for the study. Twelve different concentration combinations of 2, 4-D and kinetin were used to determine the effect of hormone concentrations on callus induction. Calli were transformed using Agrobacterium mediated transformation. For calli transformation Agrobacterium tumefaciens GV3101 containing pBI121 vector was used. Transformed callus were detected using GUS analysis. Efficiency of transient transformation was calculated for each variety. There was a significant difference of 12 treatments of callus induction of tested varieties except Gonabaru. Kaluheenaty, Madael, Gonabaru, Kottiyaran and H4 have shown the best callus induction for the same hormone concentration combination of 2.5mg/L 2,4-D and 0.5 mg/L kinetin. The present investigation was carried out at experimental orchard, Division of Fruit Science, SKUAST-K, Shalimar. The experiment consisted of three treatments on three cultivars of apple (Lal Ambri, Firdous and Sunhari) replicated thrice in RBD. The results revealed that foliar application of nutrients had a significant effect on growth parameters, foliar nutrient contents, physical & chemical characters, yield and quality of apple cultivars. The combined application of Ca, Mg and micro-nutrients resulted in better annual shoot growth, leaf fruit ratio, leaf area and number of leaves per shoot when compared with control and other treatments, without affecting uptake of N, P and K. Further, the leaf Ca, Mg, Zn, Mn and B uptake improved with the combined application of Ca, Mg & micro-nutrients when compared with control and other treatments.The studies revealed that the foliar application of nutrients improved the overall performance of apple cultivars with respect to qualitative and quantitative characters. Hence the combined foliar application of macronutrients and micro-nutrients along with soil application of N,P and K is recommended for the better performance of apple cultivars.

This book describes a simple way to practice host plant resistance phenomenon.Crop varieties resistant to insects are far less common than disease-resistant varieties, as plant breeders usually focusing more on disease resistance. However, if available, resistant varieties can be an effective defence tool against insect pests. But even when insect-resistant cultivars do not exist, some varieties may be less attractive to pest species or may tolerate more damage than others. Plant size, shape, coloration, leaf hair, cuticle thickness, and natural chemicals (attractants and repellents) can all affect pest susceptibility. Farmers can do their own breeding by collecting non-hybrid seed from healthy plants in the field. Plants well adapted to local conditions will be more likely to resist pests.

Six groundnut (Arachis hypogea L.) cultivars i.e. Golden, BARI-2000, Australian, BARD-479, ICG-7326 and 2KCGC20 were evaluated against the attack of Odontotermes obesus in terms of percentage infestation of workers and soldiers, their damage and impact on the yield. The maximum mean populations of O. obesus workers were recorded on 2KCGC20 (30.82±2.75 workers/plot) and Australian (30.65±2.92 workers/plot), followed by BARI 2000 (19.55±2.51 workers/plot), Golden (17.30±2.27 workers/plot) and BARD-479 (16.82±2.06 workers/plot). The minimum mean population of O. obesus workers was found on ICG-7326 cultivars (14.85±1.81) workers/plot. The maximum mean population infestation of O. obesus soldiers was recorded on 2KCGC20 cultivar (4.88±0.25 soldiers/plot) followed by Australian (4.15±.30, soldiers/per plot), BARD-479 (2.00±.17 soldiers/plot), ICG-7326 (1.70±.14 soldiers/plot), BARD 2000 (2.40±.25 soldiers/ plot). The minimum mean population of O. obesus soldiers was found on Gold (1.87±.17 soldiers/plot). Population of O. obesus workers was positively related with the damage and negatively related the yield in all the groundnut cultivars.

There are a large number of parameters that affect the degree of expansion of these cereals, which are connected both with the compositional characteristics of the raw material and the processing conditions. The main factors which affect puffing are puffing-temperature, puffing-duration, initial moisture content of the material to be puffed and also starch content of the raw materials. The goal of this study was to review in detail the influence of the conditions of hydro-thermal treatment of rice on the characteristic of rice as well as the puffed rice.

In the long term the country was able to reduce poverty and undernourishment, because of its steady and strong economic growth.A typical household spends nearly half of its total food intake on rice. Therefore, high rice prices make especially the poor vulnerable to price increases. In order to estimate how and to which extend rice price volatility affects demand, the daily calorie intake is focused with regard to a price increase of 10 % and 30 %.Based on the assumption that people are food insecure a total calorie intake of 2,100 calories/day/person is taken into account as poverty line. Usually households consume less rice if the price is on a higher level. In Indonesia richer households have a more price-elastic demand if rice prices increases than poor households. On the contrary, those who are net consumers or producers of rice as well as governmental supported poor households have a relatively price-inelastic demand for rice.This fact is different for households which differ in income level and location. Drought is the major climatic threat that affects agriculture of arid areas. Detection and evaluating the impact of drought with traditional methods has been laborious and time consuming. Recently methods for drought monitoring are improved. This research assessed the drought conditions at temporal basis using the meteorological and agricultural parameters integrated with geo spatial technologies. Satellite based anomalies were calculated for NDVI, rainfall and crop yield, reference evapotranspiration (ETo) evaluate comparatively the per cent change in different years. The Standardized precipitation Index was also used as an indicator. The Spatial Auto correlation regression analysis was preformed between SPI, NDVI, rainfall anomalies, total seasonal rainfall, crop yield and ET o to observe the temporal pattern of drought. NDVI anomalies and crop yield anomalies have correlated to assess the agricultural drought, NDVI and rainfall anomalies were correlated to determine the meteorological drought. Relationship between crop yields, NDVI, SPI, rainfall anomalies was used to determine the drought condition related with rainfall deficiency.

This book describes the climatic conditions that give rise to droughts, and their various forms and chief attributes. Past droughts are described including those that had severe impacts on human societies. As a disturbance, droughts can be thought of as “ramps” in that they usually build slowly and take time to become evident. As precipitation is reduced, flows from catchments into aquatic systems decline. As water declines in water bodies, ecological processes are changed and the biota can be drastically reduced, though species and populations may survive by using refuges. Recovery from drought varies in both rates and in degrees of completeness and may be a function of both refuge availability and connectivity. For the first time, this book reviews the available rather scattered literature on the impacts of drought on the flora, fauna and ecological processes of aquatic ecosystems ranging from small ponds to lakes and from streams to estuaries. The effects of drought on the biota of standing waters and flowing waters and of temporary waters and perennial systems are described and compared. In addition, the ways in which human activity can exacerbate droughts are outlined. In many parts of the world especially in the mid latitudes, global warming may result in increases in the duration and intensity of droughts. Experiments have been conducted under laboratory conditions to asses the wear behaviour of the rice husk ceramic composites in polymer matrix. From these experimental result we can also draw a fair amount of conclusion about their mechanical behaviours also i.e under stress and tensile test, hardness test etc.

Molecular markers especially, microsatellite or simple sequence repeats (SSRs) was the marker aided selection (MAS) technique utilized to determine salinity tolerance in rice. It accelerates rice breeding process by significantly improving selection efficiency. Fourty varieties were used to study the effect of salt stress on seedling growth and efficiency of SSR markers for the screening of salt tolerant varieties. Phenotyping of the varieties at the seedling stage was done using salinized (EC=12ds/m) culture solution under controlled conditions at the Bangladesh Institute of Nuclear Agriculture (BINA) glasshouse. Salt tolerant varieties showed stunted growth with the reduction of leaf area over the varieties of non-saline conditions. The seedlings grown from the normal seeds were vigorous than the seedlings from the seeds of saline conditions. This result indicated the impediment of seed vigor due to salt stress.

This study was conducted to assess the socioeconomic status and gender role of cereal crop production in two VDCs of Arghakhachi district. A pre-tested semi-structured questionnaires were used to collect the information from the respondents. Two VDCs were selected purposively and 60 HHs (30 from each VDC) were selected randomly for the survey. Cereal crop production has great contribution in HH income and livelihood upliftment. The study area was male dominated. Agriculture was the major occupation in the study area. The average area under rice (7.46 ropani) was more followed by wheat (4.58 ropani) and maize (4.35 ropani) in the study area. Hybrid variety of rice, HYV of maize and local variety of wheat was most popular. Seed was higher for wheat (10.73 kg) followed by rice (8.75 kg) and maize (6.5 kg).Total production of rice was higher than maize and wheat.The major problems in the study area were lack of technical assistance, lack of irrigation facilities and low quality seed.

Landsat Multi Spectral Scanner, Thematic Mapper, Enhanced Thematic Mapper and Nigeriasat-1satellite imagery were used to determine the rate, trend and magnitude of land use/cover changes between 1972 and 2013. A supervised classification approach using ILWIS 3.3 version was used. Four research questions were developed in line with the purpose of the study, and four correspondent hypotheses were formulated to guide the study. Relevant literatures were reviewed to cover the variables under investigation. The population of the study was 100 registered farmers in the local government area. Purposive sampling technique was used to select the population from 11 political wards of Sardauna. Data for the study were obtained by the use of a questionnaire at 0.05 level of significance and 99 degree of freedom. From the analysis, it was found that flood, desertification, land degradation, cessation of rain and drought are the major indicators of climate change in Nigeria. Insufficient extension services, lack of farmers enlightenment, negligence of government, religious/traditional believes of farmers and low farmers income are found to be major reasons the farming communities are highly vulnerable to climate change. Based on the above findings, conclusion, recommendations and suggestions for further studies were made.

Cotton is a main cash crop of Pakistan as it plays a pivotal role in the economy. Cotton plant has been genetically modified to contain the toxic gene of Bacillus thuringiensis known as Transgenic Bt Cotton and it has been found successful in the management of bollworms; however, it also invites other insect pests especially sucking pests due to reduction in pesticide sprays at early stages. So, the present study was planned to measure the comparative resistance of six Bt cotton cultivars against major sucking insect pests (whitefly, jassid and thrips). The results of experiment showed that the transgenic cotton cultivar Bt-802 found comparatively resistant by having maximum impact on whitefly and jassid abundance which contained 7.97, 1.83 per leaf population respectively. Transgenic cotton cultivar Bt-806 had maximum impact on thrips population by holding lowest population (5.80 per leaf thrips). The sucking insect population had negative correlation with relative humidity and rainfall while temperature had positive correlation. In case on psysico-morphic plant character gossypol glands and hair leaf density had significant impact on sucking insect pest population.

Growth, development and yield of soybean are a result of a variety's genetic potential interacting with environment and farming practices. Experiments were carried out in two locations for determining the resistance of some soybean genotypes to the cotton leaf worm. Beside that, identification of molecular genetic markers linked to insect resistance was also done. An attempt was done to get some new resistant hybrids through execution hybridization between these cultivars. Plant height, numbers of branches and pods per plant and seed yield per plant were decreased significantly by the infestation of cotton leaf worm (Spodoptera littoralis) as compared with soybean plants grown in Giza region (non - stressed treatment). The results of SDS-PAGE revealed a total number of 21 bands with molecular weight (MW) ranging form about 319.63 to 15.568 KDa. 5 primers showed specific ISSR markers for cotton leaf worm with 6 positive markers for cotton leaf worm resistance appeared in the resistant genotypes, while 2 negative markers for susceptibility for cotton leaf worm were present in the susceptible genotypes.

The general objective of this study is to examine the efficiency, and the factors that influence the efficiency of irrigated rice farmers in Mali. A special focus is devoted to the quantifying effects of irrigation scheme rehabilitation. A stochastic parametric frontier analysis is employed using a cross-sectional data set consisting 147 households that are cultivating 398 rice plots. It is found that for rehabilitated areas there is more room to increase rice production by making a better use of the existing technology. For these villages policies should be directed to increase the productivity of manual-labor through extension education or labor-saving technologies. On the other hand, farmers in non-re­habilitated villages are relatively more efficient indicating that there is more need for technology improvements, such as improvements in irrigation infrastructure that can enhance productive use of seed and nitrogen. A clear message of the study is that the extension services should be directed separately to heterogeneous areas. It is necessary to identify the structural differences in production environments between the areas that are located in the same region.

Rhm51 is a single copy gene, and is constitutively expressed at low levels during the cell cycle and at higher levels when the pathogen is grown in media supplemented with DNA damaging agents. We disrupted Rhm51 in two different strains of M. oryzae and both mutants showed reduced growth with conidia and colonies being highly sensitive to DNA double strand break-inducing agents. In addition, they lost the ability to undergo homologous recombination in adeA gene targeting. Mutants showed reduction in conidiation compared to the wild-type strains. Although mutants infected compatible rice cultivars, they produced significantly smaller and fewer lesions compared to wild-type strains and this might be due to the reduction in the amount of appressoria formed. The optimum level of ABA is maintained by the balance between biosynthesis and catabolism. ABA catabolism is mainly carried out by 8'-hydroxylation pathway in plants. In Arabidopsis, a Cytochrome P450 family namely CYP707A was found to be involved in the process of ABA catabolism and the present study was conducted to identify the members of CYP707A family in rice and the candidate P450 involved in ABA accumulation in developing seeds. Pattern of ABA accumulation was studied and it reaches maximum at 25 DAF. Using qualitative research methods, reviewing key publications and NGOs sites, this study identifies factors that hinder multilateral agencies, and NGOs, from delivering hygiene education to local communities in Somalia. Findings included that poor hygiene practices are more frequent in rural than in urban areas and that illiteracy, intertribal conflicts, cultural and traditional beliefs, hinder hygiene education and aid delivery to the local communities. The study recommends that training of households on contamination avoidance, hygienic water and food handling as well as water purification techniques, be part of good hygiene and practice guidelines for multilateral agencies and NGOs.

This study explored the impact of pastoral women's engagement in Income Generating Activities on gender roles taking female firewood sellers in Shinile District, Somali Region of Ethiopia as a case study. Drought dwindled away the livestock of pastoralists in the research area, undermining the traditional role that pastoral men play in food provision. Consequently, pastoral women get engaged in non-pastoral income generating activities, specifically firewood sale, for their household survival. Results indicated that women's burden increased due to their engagement in firewood trade. Also, the activity undermined women's participation in community activities as they spent a substantial amount of time on firewood collection and selling. However, despite the usual lower involvement of men in reproductive activities; their participation in domestic tasks has increased commencing up on women's engagement in firewood selling. The income gained enabled women to be a co-bread winner, even if, it is not big enough to improve their wellbeing. It is also emphasized in this study that gender roles are subject to change, for instance depending on time and one's livelihood set up.

Lr26 is also a leaf rust resistant gene found in various wheat varieties, having size of 1000 bps. The wheat varieties studied in this research was collected from various locations of Pakistan and observed that those varieties among the collected having Lr26 gene were not only resistant to the disease but also high in yield production and good in other morphological characters.
